Is the brand new PTU a series 2? Do you have a adapter sub-harness? These are known for corrosion in the connectors. Most prefer to solder the adapter harness directly into the EFI harness. Series 2 PTU on the right

When was the last time it ran properly? After the timing belt job? Since we know it is a single cylinder misfiring, we need to look for proper fuel and spark. A compression test also helps. Can you check that? Swap a coil pack with one of the other good cylinders and see if the misfire follows the coil pack. You can perform a power balance test yourself by unplugging the coil pack connectors one at a time with the engine idling. The bad cylinder will not drop in rpm as much as the others. Which is where the cylinder 2 14rpm drop came from. Conzult or another diagnostic software would go along way in your diagnosis. Especially when it comes to setting the IACV up, checking timing and TPS.
